Thermo Fisher launches the first triple LC-MS Thermo Fisher Scientific (Hereinafter referred to as: Thermo Fisher Scientific) has launched a new "triple play" system, the Thermo Scientific Orbitrap Fusion Tribrid Liquid Chromatography Mass Spectrometer (LC-MS). The system integrates quadrupole, Orbitrap and linear ion trap mass analyzers to provide unmatched depth of analysis for complex biological samples.

The new Orbitrap Fusion LC-MS debuted at the 154th Minneapolis Convention Center at the American Mass Spectrometry Annual Meeting (ASMS) held from June 9th to 13th.

“Our mission is to create the highest performance commercially available mass spectrometers,” said Dr. Ian Jardine, CTO of Thermo Fisher Scientific. “In addition, we hope that this powerful tool can be widely used in the scientific community to ensure that the instrument is easy to use. When designing Orbitrap Fusion, we integrated quadrupole, linear ion trap, and Orbitrap technology in a flexible, research-grade mass spectrometry system and improved its performance, resulting in a new generation of mass spectrometers that could revolutionize customer research."

Dr. Steven Gygi, professor of cell biology at Harvard Medical School, said: “Our biggest challenges are sensitivity and analytical throughput. The innovative Orbitrap Fusion instrument has much more quantitative accuracy than ever before and can help us achieve a broader proteomics. Coverage."

One way that the Orbitrap Fusion system solves the flux problem is in series with a mass tag (TMT). This technology enables the mass spectrometer to simultaneously perform relative quantification of proteins in multiple samples. Compared to previous tools, the Orbitrap Fusion instrument significantly improves the depth and quality of data analysis and significantly improves TMT results. The new platform uses MS3 selectivity to increase quantitative accuracy. Compared with previous analysis systems, it can obtain twice the number of MS3 scans per unit time, and the sensitivity can be significantly improved. Users can also order new TMT reagents from Thermo Fishery for up to 10 samples simultaneously.

The heart of the Orbitrap Fusion LC-MS is the configuration of three different mass analyzers. These analyzers work together to take analysis performance to new heights and bring new experimental methods:

The quadrupole is used for the parent ion selection, with a resolution of up to 0.4 amu, with excellent sensitivity and selectivity;

Super Field Orbitrap offers over 450,000 resolutions and scan rates up to 15Hz, with unparalleled analysis selectivity and speed;

Multi-pole ion cyclotron channels and dual-pressure linear ion traps provide MSn HCD, CID, and ETD cleavage, enabling fast, sensitive mass analysis at scan rates up to 20 Hz. Synchronous precursor selection also enhances the signal-to-noise ratio of the instrument.

With a three-in-one configuration, users will be able to identify more low-abundance proteins faster than existing commercial instruments. The unique instrument structure enables simultaneous parent ion isolation, cracking, and data acquisition in Orbitrap and linear ion trap mass analyzers. Compared with existing instruments, the quality of the collected data is higher, thereby extending the scope of the experiment.

Thanks to the ability to select different cleavage modes at any stage of the MSn analysis, combined with the ability of the Orbitrap or linear ion trap analyzers to detect, this enables a series of novel experiments, from which metabolites, polysaccharides, and post-translational A whole new level of structural information on modification and sequence polymorphism.

In typical metabolomics experiments, researchers often face unknowns with the presence of target compounds. To identify the unknown, the sample must be rerun on the LC-linear ion trap instrument to obtain the MSn data, but it is difficult to match the chromatographic retention time during the second run, which leads to uncertainty. But with its unique triple-analyzer configuration, the new Orbitrap Fusion Tribrid LC-MS has successfully overcome this problem, and has also provided users with the ability to innovate numerous small-molecule experimental methods with proven unknowns.

Performance is easy to implement The new intelligent software of Orbitrap Fusion LC-MS provides Dynamic Scan Management (DSM), which can automatically adjust the scan parameters with the experiment to obtain the best results.

The Orbitrap Fusion system integrates a new, easy-to-use drag-and-drop method editor to avoid guessing the vast majority of parameters in creating complex methods. The editor is part of the MS software suite and the entire suite provides unmatched applicability to the platform, including:

Thermo Scientific Freestyle Software: A new data visualization tool that also facilitates rapid method development and data quality assessment.

Thermo Scientific Proteome Discoverer: A comprehensive software program for protein identification.

Thermo Scientific Compound Discoverer Software: Used to perform small molecule structure recognition in a wide range of applications.

Thermo Scientific SIEVE Software: Differential Quantitative Analysis for Proteomics and Small Molecule Samples.

mzCloud software: A new mass spectral library that supports unknown identification and structural analysis.
